HitGen's Targeted Protein Degradation Platform integrates DEL, PROTAC synthesis, and biological validation to address historically undruggable targets.

Protein Degradation
HitGen use DEL technology to identify small molecule ligands with affinity, where DNA tags provide connection sites for linker attachment.
E3 Ligases and Their Binders
HitGen is working on identifying novel E3 ligases, and expanding the E3 ligase and binder tool box to be used for both external and internal TPD projects.
Discovery of Molecular Glue Degraders
Several dozens of purified E3s have been expressed, purified and screened against HitGen DELs, yielding many specific binders especially for those E3s without any binders reported. Some of these E3 binders have been found with potential molecular glue function.
Discovery of Proteolysis-targeting chimeras (PROTACs)
HitGen has successfully screened 41 target types. To shorten the linker optimization process, HitGen has developed a ternary complex-based PROTAC-DEL screening technology and a high-throughput PROTAC synthesis platform leveraging diverse linker repositories.
Protein Degrader Evaluation
HitGen possesses extensive expertise in biochemistry, biophysics, cell biology, in vivo pharmacokinetics, and pharmacodynamic evaluation. Given the characteristics of protein degradation technologies, HitGen has developed a series of relevant assays to support the development of protein degraders.
